研究專長: 分子細胞生物學、蛋白質運輸、細胞骨架調控

研究方向:

細胞遷移在胚胎發育、免疫監視和傷口癒合等過程扮演重要的角色。細胞的定向遷移(directed migration),通常由細胞外部的化學或機械信號所引起,需要囊胞運輸 (vesicular trafficking) 和細胞骨架重組在空間和時間上精確的協調作用。已知許多分子參與調控產生和維持細胞定向遷移。雖然許多個別分子機制已經闡明,但是囊胞運輸和細胞骨架重組之間如何協調的機制尚不清楚。小分子鳥糞嘌呤核苷酸結合蛋白 (small GTPase) 腺嘌呤核苷二磷酸核糖化因子 (ADP-ribosylation factors; Arf) 和他們的調控蛋白鳥糞嘌呤核苷酸交換因子 (guanine nucleotide-exchange factors, GEF) 可能扮演了之間的聯繫的角色。  

鳥糞嘌呤核苷酸交換因子BIG1和BIG2, 經由其Sec7 domain促使Arf與鳥糞核苷三磷酸(GTP)結合,而參與了囊胞運輸或是活化了修飾磷質脂的酵素。除了位於蛋白序列中央的Sec7 domain,在BIG1和BIG2蛋白的氨基端和羧基端還有幾個domains功能尚不清楚。BIG2基因突變已被發現會造成人類隱性遺傳的腦室周圍白質異位(periventricular heterotopia)和小頭畸形(microcephaly)。我們之前的研究發現BIG1和BIG2可調控肌動蛋白而影響細胞遷移。使用siRNA減少細胞中BIG1或BIG2的表現,會增加纖維狀肌動蛋白的含量,減少細胞膜突起而影響細胞移動。然而BIG1和BIG2的上游調節分子和信號,以及如何通過BIG1,BIG2來調控細胞移動的分子機制仍不清楚,我們將結合細胞生物學,分子生物,生化和蛋白質體學的技術來鑑定和探討BIG1和BIG2蛋白質複合體在囊胞運輸和細胞遷移過程中的功能,以及Arf在這些過程中扮演的角色。目標是找出BIG1和BIG2參與囊胞運輸和細胞遷移的分子調控機制。所獲得的信息將使我們能夠了解因小分子鳥糞嘌呤核苷酸結合蛋白和其調節蛋白所造成的疾病,而設計對策進行分析和操縱,期能有助於疾病治療的改善。
