研究專長: 染色體損壞修復的機制、癌症基因體、酵母基因學

研究方向:

癌細胞具有基因體不穩定性的特性,而維持基因體的穩定是所有生物最重要的機制,以維持各物種可以代代遺傳下去。我們的研究主要在探討細胞如何維持基因體的穩定性,以及為何癌症會失去此一穩定性,基因體的穩定是由哪些基因所維持。因此我們除了利用模式生物酵母並與成大醫院腫瘤醫學部合作,由提供的腫瘤樣本,利用比較基因體找出癌細胞中變異的基因,並且透過分子生物學與基因體學,來探究此基因的功能。長期目標以找出致癌基因成癮(oncogene addiction)的路徑,作為標靶治療以及實現個人化醫學(personalized medicine)。

目前研究主要著重在染色體修復路徑,post-replication repair與組蛋白修飾(histone modification),在染色體穩定性所扮演的角色。

 

 

環境中的毒物或是細胞代謝過程中所產生的過氧化物,會對我們的DNA造成損壞,危害我們基因的穩定性。這些被損壞的DNA會造成細胞在複製的過程中的障礙,造成DNA複製停滯的現象(stalled DNA replication),長期的停滯,最終會導致DNA雙股斷裂(DNA double-strand breaks),以及細胞死亡。為了要防止這種情況的發生,細胞演化出後複製修復(post-replication repair)的救命機制,來克服在DNA複製過程中所遭遇的DNA損壞。我們最近的研究顯示許多參與後複製修復的基因在防止細胞癌化上,扮演很重要的角色。
